Mix the culture medium into the tank as needed, seal it as required, and then put the small fermentation tank (5L) into a large sterilization pot for sterilization (115 ℃, 30 minutes); Large and medium-sized fermentation tanks (10L and above) are sterilized with steam (121 ℃, 30 minutes).
3. After the tank has cooled down, place it on the fermentation platform and install it properly; Turn on the cooling water, turn on the power supply of the air pump, connect the ventilation pipe to start ventilation, adjust the intake knob to adjust the ventilation amount appropriately, and maintain the tank pressure at 0.05MPa; Set temperature, pH, stirring speed, etc., and calibrate the slope of the dissolved oxygen electrode at a determined speed and ventilation rate, with a value of 100%
4. After the temperature stabilizes and all parameters are correct, connect the pre shaken seeds, start fermentation timing, and start recording various parameters.
5. After fermentation, clean the tank and electrodes, insert the pH electrode into a triangular bottle containing 3M potassium chloride for later use, and cover the probe of the dissolved oxygen electrode with a protective sleeve. Store for later use.
